Abstract

Hundreds post-translational modifications (PTM) were characterized among which a large variety of glycosylations including O-GlcNAcylation. Since its discovery, O-GlcNAcylation has emerged as an unavoidable PTM widespread in the living beings including animal and plant cells, protists, bacteria and viruses. In opposition to N- and O-glycosylations, O-GlcNAcylation only consists in the transfer of a single N-acetylglucosamine moiety through a beta-linkage onto serine and threonine residues of proteins confined within the cytosol, the nucleus and the mitochondria. The O-GlcNAc group is provided by UDP-GlcNAc, the end-product of the hexosamine biosynthetic pathway located at the crossroad of cell metabolisms making O-GlcNAcylation a PTM which level tightly reflects nutritional status; therefore regulation of cell homeostasis should be intimately correlated to lifestyle and environment. Like phosphorylation, with which it can compete, O-GlcNAcylation is reversible. This versatility is managed by OGT (O-GlcNAc transferase) that transfers the GlcNAc group and OGA (O-GlcNAcase) that removes it. Also, like its unsweetened counterpart, O-GlcNAcylation controls fundamental processes, e.g. protein fate, chromatin topology, DNA demethylation and, as recently revealed, circadian clock. Deregulation of O-GlcNAc dynamism may be involved in the emergence of cancers, neuronal and metabolic disorders such as Alzheimer's or diabetes respectively. Abstract

Escherichia coli is a versatile organism and very diverse. Members of this species vary from very pathogenic agents causing different types of diseases including meningitis, gastroenteritis, and septicemia, just to cite a few, to harmless organisms living in the intestines of both humans and animals. E. coli has also been used as a model organism for most bacteria except a few. For this reason, its study provides a huge advantage and can help understand the mechanisms involved in different processes such as pathogenesis, environmental disinfection, nutrient utilization, antibiotic resistance, and diagnostic/detection methods, and these are indeed the topics discussed in this book. The book has been divided into four main sections representing the different facets of E. coli applications, which include disease, biotechnology, environmental engineering and innovative approaches to detection, and lastly its physiology and cell biology. Such processes can be applied to the study of other organisms as well considering the development of diversity; for example, many organisms are capable of horizontal gene transfer, which is capable of increasing the fitness of the bacterial organisms involved and has a great impact on the control of such bacterial organism.

Abstract

A significant increase in the prevalence of campylobacteriosis cases has been observed over the past years. Campylobacter has emerged as the leading cause of bacterial foodborne disease worldwide with a significant impact on human health and an associated economic burdens. Campylobacteriosis human cases have been generally correlated with the handling, preparation and consumption of poultry. In 2017, the European Commission regulation has amended Regulation (EC) No 2073/2005 on the hygiene of foodstuffs as regards Campylobacter on broiler carcasses stating a limit of 1000 cfu/g. Campylobacter is also present in other farm animals and is frequently found on a range of foodstuffs due to cross contamination. Among the pathogenic species, C. jejuni is the most prevalent species followed by C. coli. Current guidelines highlight the importance of biosecurity but these measures are failing to mitigate the risk of pathogenic Campylobacter. As an obligate microaerophile, Campylobacter does not multiply under atmospheric oxygen concentration at ambient temperatures. It therefore constitutes a puzzle as to how it can survive from farm to retail outlets. The underlying molecular mechanisms of persistence, survival and pathogenesis appear to be unique to this pathogen. Recent research has indicated how genomic polymorphism, restricted catabolic capacity, self regulation or deregulation of genes, bacterial cooperation and unknown contamination routes may be connected to this specificity.This book includes original studies on both C. jejuni and C. coli species dealing with epidemiology and animal carriage, host interaction, control strategies, metabolism and regulation specificities of these two pathogenic species, methodology to improve cultural techniques and chicken gut microbiota challenged with Campylobacter.

Abstract

By stimulating adrenal gland and corticosteroid synthesis, the adrenocorticotropic hormone (ACTH) plays a central role in response to stress. In this Research Topic, a particular attention has been given to the recent developments on adrenocortical zonation; the growth-promoting activities of ACTH; the various steps involved in acute and chronic regulation of steroid secretion by ACTH, including the effect of ACTH on circadian rhythms of glucocorticoid secretion. The Research Topic also reviews progress and challenges surrounding the properties of ACTH binding to the MC2 receptor (MC2R), including the importance of melanocortin-2 receptor accessory protein (MRAP) in MC2R expression and function, the various intracellular signaling cascades, which involve not only protein kinase A, the key mediator of ACTH action, but also phosphatases, phosphodiesterases, ion channels and the cytoskeleton. The importance of the proteins involved in the cell detoxification is also considered, in particular the effect that ACTH has on protection against reactive oxygen species generated during steroidogenesis. The impact of the cellular microenvironment, including local production of ACTH is discussed, both as an important factor in the maintenance of homeostasis, but also in pathological situations, such as severe inflammation. Finally, the Research Topic reviews the role that the pituitary-adrenal axis may have in the development of metabolic disorders. In addition to mutations or alterations of expression of genes encoding components of the steroidogenesis and signaling pathways, chronic stress and sleep disturbance are both associated with hyperactivity of the adrenal gland. A resulting effect is increased glucocorticoid secretion inducing food intake and weight gain, which, in turn, leads to insulin and leptin resistance. These aspects are described in detail in this Research Topic by key investigators in the field. Many of the aspects addressed in this Research Topic still represent a stimulus for future studies, their outcome aimed at providing evidence of the central position occupied by the adrenal cortex in many metabolic functions when its homeostasis is disrupted. An in-depth investigation of the mechanisms underlying these pathways will be invaluable in developing new therapeutic tools and strategies.

Abstract

Adipose tissue is a specialized connective tissue which, depending on type, plays different and significant roles in the human body: protects against environmental factors, stores lipids and triacylglycerol, synthesizes fatty acids and is involved in the process of thermogenesis. It is also a major secretory organ and highly active metabolic tissue. It secretes, for example, cholesterol, retinol, steroid hormones, prostaglandins and proteins known as ""adipokines"". Some of these molecules may be associated with pathologies such as obesity and insulin resistance. In humans, there are two main sites of adipose tissue accumulation: visceral and subcutaneous. Obesity is a worldwide health problem. This book also discusses a series of up-to-date topics about this pathology.

Abstract

The adrenal gland plays essential roles in the control of body homeostasis, stress and immune responses. The adrenal cortex represents up to 90% of the gland and is specialised in the production of mineralocorticoids, glucocorticoids and adrenal androgens. This production is tightly coordinated and results from a unique zonal organisation. Although our knowledge of the molecular mechanisms controlling adrenal steroidogenesis is quite extensive, for decades, the mechanisms of adrenal cortex development, cellular homeostasis and renewal have remained elusive. The advent of new high-throughput technologies and sophisticated genetic approaches has brought tremendous progress in our understanding of how the adrenal cortex achieves and maintains its particular organisation. The aim of this Frontiers in Endocrinology Topic is to provide readers with a snapshot of our current knowledge on adrenal physiology and how deregulations of these processes result in adrenal diseases. This includes but is not limited to, basic research on adrenal development, cell lineage identification, progenitor cells, tissue renewal, control of differentiation and zonation and clinical research on the identification of disease-related genes.
