Abstract

Abstract

Since the beginning of times, pain treatment has been the motive of research giving birth to multiple groups of pharmacological families and therapies. Pain perception is a construction built over the biological phenomenon of signal transduction surrounded by different factors such as gender, age, and sociocultural status, among others. The concept of pain as the solely biological manifestation of defense is nowadays considered as a narrow-minded view of this topic. In this regard concepts such as newborns feel no pain or older people complain about everything therefore should not be paid attention when referring pain, are being left behind in the understanding that pain alleviation is a human right and everybody feeling pain should be helped for its relief. This book comprises many aspects of pain treatment and the drugs involved in it. From old analgesics with new mechanisms of action for pain alleviation to analgesics potential for diminishing oxidative stress; from pharmacological therapies to electrical ones, going through alternative medicine; and from pain treatment in dentistry to chronic pain therapies, also boarding the treatment of migraine, different experts share their knowledge on the topic.

Abstract

The scientific monograph by the author Peter Mikus entitled ""Chiral Capillary Electrophoresis in Current Pharmaceutical and Biomedical Analysis"" provides a comprehensive view on the advanced capillary electrophoresis techniques aimed to current chiral bioanalysis. The advances in the chiral electrophoresis analytical approaches are divided and theoretically described in three sections involving (i) advanced chiral separations for the optimization of chiral resolution (separation mechanisms; electrophoresis techniques in capillary and microchip format; electrophoretic modes such as ITP, CZE/EKC, CEC; chiral additives / pseudophases / phases), (ii) advanced sample preparation for the on-line preconcentration, sample clean-up and analyte derivatization (implementation of electrophoretic effects such as stacking; non-electrophoretic effects such as SPE, chromatography, dialysis; combinations of these effects; multidimensional CE systems; instrumental schemes), (iii) advanced combinations of detection and electrophoresis for the optimization in qualitative and quantitative evaluation (the most important universal as well as selective detection approaches such as absorption and fluorescence spectrophotometry, electrochemical detection, mass spectrometry vs. (i) and/or (ii)). Real analytical potential (benefits and limitations) of these advanced analytical approaches is emphasized by selected performance parameters of the methods and illustrated by many current practical applications including chiral analyses of drugs, their (bio)degradation products and biomarkers in pharmaceutical and biological matrices. The author wishes the readers many inspirations in the creation of new innovative approaches in the field of advanced chiral electrophoresis techniques with the aim to overcome capabilities of the current analytical techniques.

Abstract

Canadian Pharmaceutical Scientists have a rich history of ground-breaking research in pharmacokinetics and drug metabolism undertaken throughout its Pharmacy and Medical Schools and within the Pharmaceutical and biotechnology industry. The principle of drug Absorption, Distribution, Metabolism and Excretion (ADME) is the foundational basis of rationale drug-design, and pharmacotherapy. The study of ADME and its descriptive quantitative analysis is the basis of pharmacokinetics. Pharmacokinetics is fundamental in the development of a new chemical entity into a marketable product and is essential in understanding the bioavailability, bioequivalence and biosimilarities of drugs. Pharmacokinetics and drug development studies facilitate an understanding of organ-based functionality. Population pharmacokinetic variability and the modeling of drug concentrations has significant utility in translating individual response in a target patient population.This special issue serves to highlight and capture the contemporary progress and current landscape of pharmacokinetics and drug metabolism within the prevailing Canadian context. We invite articles on all aspects of Pharmacokinetics and Drug Metabolism studies highlighting the world-class research currently undertaken in Canada for this special issue.

Abstract

For thousands of years, pharmacological knowledge coming from natural remedies, has been handed down from generation to generation, without any awareness of the ways in which preparations are made to face diseases. The advent of pharmaceutical chemistry and of the modern drug industry turned that lack of awareness into a scientific knowledge that changed the destiny of the human race. The twenty-eight chapters of this book, are taken from the lectures held by Professor Ettore Novellino every year in his course “Pharmaceutical Chemistry and Toxicology 2”. The first chapters address the basic notions of drugs, homeostasis, pharmacopoeia, and receptor; then, the different pharmaceutical classes are introduced by analyzing their pharmacological and chemical aspects. In particular, the structural study of the interaction between drugs and receptors or biological enzymes gives the fundamentals to connect the chemical and stereochemical properties of a compound family, with the biological activity, a correlation better known as Quantitative Structure-Activity Relationship (QSAR). Several examples of the synthesis of some of the most historically renown drugs, provided at the end of each chapter, integrate the book.

Abstract
