Abstract

The role of major dietary omega-3 fatty acids (Ω-3; α-linolenic acid, eicosapentaenoic acid and docosahexaenoic acid) in human health has generated enormous scientific interest and many controversies in recent years. Due to a growing number of studies with conflicting or even negative clinical results, the former “hype” of Ω-3 thought to be beneficial in many aspects of human health regardless of the physiological and clinical preconditions is now being critically re-evaluated, especially with respect to the potential role of Ω-3 fatty acid supplementation in preventing a variety of diseases and clinical conditions. This critical view reflects the complex interaction of Ω-3 with cell membranes and their integrated proteins mediating signal transduction, transport systems, and other processes. Moreover, Ω-3 are precursors of bioactive metabolites, such as eicosanoids, lipoxins, resolvins, protectins, maresins, and nitrolipids that influence several physiological and pathophysiological processes and their full spectrum of effects are only beginning to be defined. Finally, physiological and pathophysiological conditions as well as concomitant pharmacological treatments may influence the specific and non-specific actions of Ω-3 supplementation.This Special Issue of the Journal of Clinical Medicine will emphasize the role and biological interactions of Ω-3 with regard to cancer, psychiatric disorders, metabolic disorders and nutrition and will also reflect on some basic molecular and cellular mechanisms.

Abstract

The Reeler mutation was so named because of the alterations in gait that characterize homozygous mice. Several decades after the description of the Reeler phenotype, the mutated protein was discovered and named Reelin (Reln). Reln controls a number of fundamental steps in embryonic and postnatal brain development. A prominent embryonic function is the control of radial neuronal migration. As a consequence, homozygous Reeler mutants show disrupted cell layering in cortical brain structures. Reln also promotes postnatal neuronal maturation. Heterozygous mutants exhibit defects in dendrite extension and synapse formation, correlating with behavioral and cognitive deficits that are detectable at adult ages. The Reln-encoding gene is highly conserved between mice and humans. In humans, homozygous RELN mutations cause lissencephaly with cerebellar hypoplasia, a severe neuronal migration disorder that is reminiscent of the Reeler phenotype. In addition, RELN deficiency or dysfunction is also correlated with psychiatric and cognitive disorders, such as schizophrenia, bipolar disorder and autism, as well as some forms of epilepsy and Alzheimer's disease. Despite the wealth of anatomical studies of the Reeler mouse brain, and the molecular dissection of Reln signaling mechanisms, the consequences of Reln deficiency on the development and function of the human brain are not yet completely understood. This Research Topic include reviews that summarize our current knowledge of the molecular aspects of Reln function, original articles that advance our understanding of its expression and function in different brain regions, and reviews that critically assess the potential role of Reln in human psychiatric and cognitive disorders.

Abstract

Traditionally, abnormalities of neurons and neuronal networks including synaptic abnormalities and disturbance of neurotransmitters have dominantly been believed to be the main causes of psychiatric disorders. Recent cellular neuroscience has revealed various unknown roles of glial cells such as astrocytes, oligodendrocytes and microglia. These glial cells have proved to continuously contact with neurons /synapses, and have been shown to play important roles in brain development, homeostasis and various brain functions. Beyond the classic neuronal doctrine, accumulating evidence has suggested that abnormalities and disturbances of neuron-glia crosstalk may induce psychiatric disorders, while these mechanisms have not been well understood. This Research Topic of the Frontiers in Cellular Neuroscience will focus on the most recent developments and ideas in the study of glial cells (astrocytes, oligodendrocytes and microglia) focusing on psychiatric disorders such as schizophrenia, mood disorders and autism. Not only molecular, cellular and pharmacological approaches using in vitro / in vivo experimental methods but also translational research approaches are welcome. Novel translational research approaches, for example, using novel techniques such as induced pluripotent stem (iPS) cells, may lead to novel solutions. We believe that investigations to clarify the correlation between glial cells and psychiatric disorders contribute to a novel understanding of the pathophysiology of these disorders and the development of effective treatment strategies.

Abstract

The discovery of the involvement of neuropeptides with behaviours other than regulatory motivated ones took place in the midst of 1960’s with David de Wied’s first report on the influence of pituitary peptides on memory. This major scientific breakthrough opened a new frontier of studies in Endocrinology and its related fields, Neuroendocrinology and Psychoneuroendocrinology. Neuropeptides were initially thought to be involved in homeostatic regulation and secreted only from neurons located in the hypothalamus; they are now recognized neurotransmitters, produced in and secreted from distinct brain areas, associated with a myriad of, not only, motivated, but also psychopathological behaviours. Motivated behaviours are determinant for individual and species survival, but their expression in a large spectrum and deviations from average may give rise to a number of psychiatric conditions.

Abstract

euromodulation is among the fastest-growing areas of medicine, involving many diverse specialties and affecting hundreds of thousands of patients with numerous disorders worldwide. It can briefly be described as the science of how electrical, chemical, and mechanical interventions can modulate the nervous system function. A prominent example of neuromodulation is deep brain stimulation (DBS), an intervention that reflects a fundamental shift in the understanding of neurological and psychiatric diseases: namely as resulting from a dysfunctional activity pattern in a defined neuronal network that can be normalized by targeted stimulation. The application of DBS has grown remarkably and more than 130,000 patients worldwide have obtained a DBS intervention in the past 30 years—most of them for treating movement disorders. This Frontiers Research Topics provides an overview on the current discussion beyond basic research in DBS and other brain stimulation technologies. Researchers from various disciplines, who are working on broader clinical, ethical and social issues related to DBS and related neuromodulation technologies, have contributed to this research topic.

Abstract

It is now well appreciated that the immune system, in addition to its traditional role in defending the organism against pathogens, communicate in a well-organized fashion with the brain to maintain homeostasis and regulate a set of neural functions. Perturbation in this brain-immune interactions due to inflammatory responses may lead to psychiatric and neurological disorders. Microglia are one of the essential cells involved in the brain-immune interactions. Microglial cells are now not simply regarded as resident tissue macrophages in the brain. These cells are derived from myeloid progenitor cells in the yolk sac in early gestation, travel to the brain parenchyma and interact actively with neurons during the critical period of neurogenesis. Microglia provide a trophic support to developing neurons and take part in the neural wiring through the activity-dependent synapse elimination via direct neuron-microglia interactions. Altered microglial functions including changes in the gene expression due to early life inflammatory events or psychological and environmental stressors can be causally related to neurodevelopmental diseases and mental health disorders. This type of alterations in the neural functions can occur in the absence of infiltration of inflammatory cells in the brain parenchyma or leptomeninges. In this sense, the pathogenetic state underlying a significant part of psychiatric and neurological diseases may be similar to “para-inflammation”, an intermediate state between homeostatic and classical inflammatory states as defined by Ruslan Medzhitov (Nature 454:428-35, 2008). Therefore, it is important to study how systemic inflammation affects brain health and how local peripheral inflammation induces changes in the brain microenvironment. Chronic pain is also induced by disturbance in otherwise well-organized multisystem interplay comprising of reciprocal neural, endocrine and immune interactions. Especially, early-life insults including exposure to immune challenges can alter the neuroanatomical components of nociception, which induces altered pain response later in life. Recently the discrete roles of microglia and blood monocyte-derived macrophages are being defined. The distinction may be further highlighted by disorders in which the brain parenchymal tissue is damaged. Therefore, studies investigating the dynamics of immune cells in traumatic brain injury and neurotropic viral infections including human immunodeficiency virus, etc. as well as neurodegenerative diseases such as amyotrophic lateral sclerosis are promising to clarify the interplay between the central nervous and immune systems. The understanding of the histological architecture providing the infrastructure of such neuro-immune interplay is also essential. This Frontiers research topic brings together fourteen articles and aims to create a platform for researchers in the field of psychoneuroimmunology to share the recent theories, hypotheses and future perspectives regarding open questions on the mechanisms of cell-cell interactions with chemical mediators among the nervous, immune and endocrine systems. We hope that this platform would reveal the relevance of the studies on multisystem interactions to enhance the understanding of the mechanisms underlying a wide variety of neurological and psychiatric disorders.

Abstract

Parenthood represents a fundamental construct that identifies the quality of early adult-infant interactions. In both short and long period, relationships, as primary interactional experiences, have an essential role in influencing individual’s adjustment and psychopathology development during in lifetime. In this scenario, the most important areas of interest and innovation are (a) parents’ representation of themselves, both in relation to the child and to their care-giving role; (b) the quality of couple relationship, in terms of both conjugal and co-parental bounds, and its influence on the quality of early mother-father-child interactions; (c) the early models of caregiver-baby interaction; (d) the recent approach to the “maternal brain”, that represents the contribution coming from neurosciences, linked to the adult’s activation and cerebral functioning processes, in association with the parental role. These data are a starting point for the individuation of functioning mechanisms and developmental trajectories of parenting in groups of adults and babies, belonging to normative populations. At the same time, those studies may provide an important introduction to the detection of critical and/or dysfunctional aspects in populations of babies and adults at risk (e.g. preterm babies, adopted children, etc.) or in adults’ clinical group (e.g. a depressed parents, addicted parents, etc.) or in children with one or more impaired characteristic (e.g. children with organic diseases, with autistic disorders, etc.).. The present Research Topic puts the attention on these themes, particularly, considering the possible aftermaths that empirical research may have on planning and realizing interventional models to support the parental functioning. We will consider papers focused on the individuation of methods of observation and assessment, which allow to plan and realize prevention programs and/or interventions, primarily focused on parental support, both in the early stadium of the child’s development and in the long-term period. Thus, the focus is both on the prevention and sustenance sphere and, also, on specific manners of taking on the therapeutic responsibility of the adult-infant relationship.

Abstract

Neuropsychiatric disorders have long been considered as specific dysfunctions of neuronal functions. Studies of the recent decade, however, have challenged this simplistic view, highlighting the important role played by neuroglial cells in the onset and/or progression of neuropsychiatric diseases. In the central nervous system (CNS) non-excitable neuroglia are represented by cells of ectodermal origin (astrocytes, mainly responsible for CNS homeostasis and oligodendrocytes that provide myelination and support for axons) and mesodermal origin (microglial cells that are scions of foetal macrophages entering the neural tube early in development; these cells provide for CNS defence and contribute to shaping neuronal networks). Pathological changes of neuroglia are complex; these changes are classified into reactive gliosis (astrogliosis, activation of microglia and hypertrophy of oligodendroglial precursors), gliodegeneration with loss of function and glial pathological remodelling. Combination of these processes defines the evolution of neurological diseases in general and neuropsychiatric disorders in particular.In this research topic we addressed the contribution of neuroglia to major neuropsychiatric pathologies including major depression, schizophrenia, and addictive disorders.
