Abstract

The role of major dietary omega-3 fatty acids (Ω-3; α-linolenic acid, eicosapentaenoic acid and docosahexaenoic acid) in human health has generated enormous scientific interest and many controversies in recent years. Due to a growing number of studies with conflicting or even negative clinical results, the former “hype” of Ω-3 thought to be beneficial in many aspects of human health regardless of the physiological and clinical preconditions is now being critically re-evaluated, especially with respect to the potential role of Ω-3 fatty acid supplementation in preventing a variety of diseases and clinical conditions. This critical view reflects the complex interaction of Ω-3 with cell membranes and their integrated proteins mediating signal transduction, transport systems, and other processes. Moreover, Ω-3 are precursors of bioactive metabolites, such as eicosanoids, lipoxins, resolvins, protectins, maresins, and nitrolipids that influence several physiological and pathophysiological processes and their full spectrum of effects are only beginning to be defined. Finally, physiological and pathophysiological conditions as well as concomitant pharmacological treatments may influence the specific and non-specific actions of Ω-3 supplementation.This Special Issue of the Journal of Clinical Medicine will emphasize the role and biological interactions of Ω-3 with regard to cancer, psychiatric disorders, metabolic disorders and nutrition and will also reflect on some basic molecular and cellular mechanisms.

Abstract

The high number of papers submitted and ultimately accepted for publication in this special issue attests the great amount of research being conducted on TSPO and its role in living cells. Thus, TSPO has become an extremely attractive subcellular biomark for the early detection of disease states overexpressing this protein and for the selective delivery to mitochondria of drugs and probes. Moreover, the effort in the design and synthesis of new, more specific and effective TSPO ligands proves to be very valuable. All these topics have been addressed in the special issue.

Abstract

Alzheimer’s disease (AD) is an age-related neurological disease that affects tens of millions of people, in addition to their carers. Hallmark features of AD include plaques composed of amyloid beta, as well as neurofibrillary tangles of tau protein. However, despite more than a century of study, the cause of Alzheimer’s disease remains unresolved. The roles of amyloid beta and tau are being questioned and other causes of AD are now under consideration. The contributions of researchers, model organisms, and various hypotheses will be examined in this Special Issue.

Abstract

Networks of coordinated interactions among biological entities govern a myriad of biological functions that span a wide range of both length and time scales—from ecosystems to individual cells and from years to milliseconds. For these networks, the concept “the whole is greater than the sum of its parts” applies as a norm rather than an exception. Meanwhile, continued advances in molecular biology and high-throughput technology have enabled a broad and systematic interrogation of whole-cell networks, allowing the investigation of biological processes and functions at unprecedented breadth and resolution—even down to the single-cell level. The explosion of biological data, especially molecular-level intracellular data, necessitates new paradigms for unraveling the complexity of biological networks and for understanding how biological functions emerge from such networks. These paradigms introduce new challenges related to the analysis of networks in which quantitative approaches such as machine learning and mathematical modeling play an indispensable role. The Special Issue on “Biological Networks” showcases advances in the development and application of in silico network modeling and analysis of biological systems.

Abstract

The contribution of genomic variants to the aetiopathogenesis of both paediatric and adult neurological disease is being increasingly recognized. The use of next-generation sequencing has led to the discovery of novel neurodevelopmental disorders, as exemplified by the deciphering developmental disorders (DDD) study, and provided insight into the aetiopathogenesis of common adult neurological diseases. Despite these advances, many challenges remain. Correctly classifying the pathogenicity of genomic variants from amongst the large number of variants identified by next-generation sequencing is recognized as perhaps the major challenge facing the field. Deep phenotyping (e.g., imaging, movement analysis) techniques can aid variant interpretation by correctly classifying individuals as affected or unaffected for segregation studies. The lack of information on the clinical phenotype of novel genetic subtypes of neurological disease creates limitations for genetic counselling. Both deep phenotyping and qualitative studies can capture the clinical and patient’s perspective on a disease and provide valuable information. This Special Issue aims to highlight how next-generation sequencing techniques have revolutionised our understanding of the aetiology of brain disease and describe the contribution of deep phenotyping studies to a variant interpretation and understanding of natural history.

Abstract

The role of nutrition in health and disease has been appreciated from time immemorial. Around 400 B.C., Hippocrates wrote “Let food by thy medicine and medicine be thy food.” In the 12th century, the great philosopher and physician Moses Maimonides wrote “any disease that can be treated by diet should be treated by no other means.” Now, in the 21st century, we are bombarded by claims in the media of “superfoods,” wondrous nutritional supplements, and special diets that promise to cure or prevent disease, improve health and restore functioning. Much of the focus has been on neurological disease, brain health and psychological functioning (behavior, cognition, and emotion). The hyperbole aside, there has been considerable progress in the past decade in our understanding of the contribution of specific nutrients and dietary patterns to brain development, physiology, and functioning. This Special Issue of Brain Sciences is devoted to the latest research on the role of nutritional deficiencies and excesses in the genesis of brain dysfunction, and use of diet for the prevention and treatment of brain and mental disorders. Basic laboratory and clinical research studies of the immature, adult, and aged nervous system are all welcome.

Abstract

In this special issue of Diagnostics, expert contributors have produced up-to-date research studies and reviews on various topics related to the diagnosis of dementia and cognitive impairment. The methods of the assessments discussed extend from simple neurological signs, which may be elicited in the clinical encounter, through cognitive screening instruments, to sophisticated analyses of neuroimaging and cerebrospinal fluid biomarkers of disease. It is hoped that these various methods may facilitate earlier diagnosis of dementia and its subtypes, and provide differential diagnosis of depression and functional cognitive disorders, as a prelude to meaningful interventions.

Abstract

The range of human neurodegenerative diseases continues to pose significant unmet medical needs for societies around the world. The progressive and terminal nature of these conditions places a considerable personal burden on the individual affected but also on public health systems and health services. Tens of millions of people are indiscriminately affected by various dementias, which are rising at an alarming rate. There are no cures for many conditions, and it is clear that treatments applied as early as possible could greatly improve outcomes for patients. Therefore, new disease classification and diagnostic tools should be a key priority. Metabolomics represents a relatively new field of analytical science, which can be extremely useful in the early diagnosis of disease. The relatively unique feature of metabolites is that they sit at the intersection between the genetic background of an organism and its environment. Because many neurodegenerative diseases are not genetically inherited (instead having a range of known genetic risk factors and also a large number of unknown environmental triggers) the field of metabolomics offers great promise for the discovery of new, biologically, and clinically relevant biomarkers for neurodegenerative disorders. It is already bringing forward new knowledge in terms of the mechanisms of neurodegenerative disease.

Abstract

While there is a tremendous literature on the topic of wine and health ranging back to the days of Hippocrates, it is considered that there is an unlimited variety of wine, allowing for the association of senses, nutrition, and hedonism. The history of vine and wine has lasted for at least 7000 years. Vitis represent adaptable plants, and thanks to the large variety of strains, wine is an alchemical mix with unique properties, a rich and original composition in terms of polyphenols, and well known antioxidants. This explains why wine and health are closely linked to nutrition.

Abstract

The Neuroproteomics Special Issue overviews the unique challenges that must be addressed to carry out meaningful MS/proteomics analyses on neural tissues and the technologies that are available to meet these challenges. The articles on Alzheimer’s disease, addiction, and schizophrenia illustrate how MS/proteomics technologies can be used to improve our ability to diagnose and understand the molecular basis for neurological diseases. Several articles will be of interest to investigators beyond the field of neurological disorders. The review on the discovery of biofluid biomarkers of neurodegenerative dementias will be of interest to investigators searching for other disease biomarkers. Similarly, the review on the role of neuroproteomics in elucidating mechanisms of drug addiction provides an overview of the utility of MS/proteomics approaches for addressing critical questions in addiction neuroscience that should be applicable to investigators involved in virtually any area of biomedical research. Likewise, the article on developing targeted MS approaches for quantifying postsynaptic density proteins will be useful for any investigator who wishes to design targeted assays for virtually any protein. Finally, the peroxidase-mediated proximity labeling technology, described in the article on mapping the proteome of the synaptic cleft, will be of interest to investigators interested in mapping other spatially restricted proteomes.
