Abstract

Tissue engineering and regenerative medicine is a rapidly evolving research field which effectively combines stem cells and biologic scaffolds in order to replace damaged tissues. Biologic scaffolds can be produced through the removal of resident cellular populations using several tissue engineering approaches, such as the decellularization method. Indeed, the decellularization method aims to develop a cell-free biologic scaffold while keeping the extracellular matrix (ECM) intact. Furthermore, biologic scaffolds have been investigated for their in vitro potential for whole organ development. Currently, clinical products composed of decellularized matrices, such as pericardium, urinary bladder, small intestine, heart valves, nerve conduits, trachea, and vessels, are being evaluated for use in human clinical trials. Tissue engineering strategies require the interaction of biologic scaffolds with cellular populations. Among them, stem cells are characterized by unlimited cell division, self-renewal, and differentiation potential, distinguishing themselves as a frontline source for the repopulation of decellularized matrices and scaffolds. Under this scheme, stem cells can be isolated from patients, expanded under good manufacturing practices (GMPs), used for the repopulation of biologic scaffolds and, finally, returned to the patient. The interaction between scaffolds and stem cells is thought to be crucial for their infiltration, adhesion, and differentiation into specific cell types. In addition, biomedical devices such as bioreactors contribute to the uniform repopulation of scaffolds. Until now, remarkable efforts have been made by the scientific society in order to establish the proper repopulation conditions of decellularized matrices and scaffolds. However, parameters such as stem cell number, in vitro cultivation conditions, and specific growth media composition need further evaluation. The ultimate goal is the development of “artificial” tissues similar to native ones, which is achieved by properly combining stem cells and biologic scaffolds and thus bringing them one step closer to personalized medicine. The original research articles and comprehensive reviews in this Special Issue deal with the use of stem cells and biologic scaffolds that utilize state-of-the-art tissue engineering and regenerative medicine approaches.

Abstract

The aim of this Special Issue is to review, understand, and evaluate new and exciting opportunities from the field on regenerative medicine, biomaterials, and stem cell research for the bioengineering of human liver grafts that can be applied for transplantation and personalized treatment of end-stage liver disease.The development of culture conditions for long-term expansion of LGR5+ intestinal stem cells as crypt-villus structures demonstrated the feasibility of deriving complex, organ-like structures in vitro from primary adult tissues, including the liver. Moreover, human pluripotent stem cells (hPSCs) can be applied to generate functionally maturated liver and bile duct epithelial cells.In this Special Issue, we welcome reviews and original papers focussing on hepatic cell sources, including adult hepatic stem cells, organoids, fetal and induced pluripotent stem cells, and primary cells (i.e., hepatocytes, cholangiocytes, and endothelial cells) and how these cells can be applied in tissue engineering strategies to generate implantable and personalized liver grafts. Potential topics include, but are not limited to, the following: liver tissue engineering, liver regeneration, graft repair, liver stem cells and organoids, bio-scaffolds, and 3D printing.We invite you to contribute original research papers, as well as comprehensive reviews, aligned with these themes, to advance and improve the actual state-of-the-art in liver bioengineering and providing new opportunities for the imminent medical problem of organ and tissue shortage for transplantation.

Abstract

This book focus principally on ions-releasing and other smart dental materials for application in preventive and restorative dentistry, as well as in endodontics in the form of adhesives, resin-based composites, pastes, varnishes, liners and dental cements. Special attention has been given to bioactive materials developed to induce cells differentiation/stimulation, hard tissue formation and exert antimicrobial actions. New innovations are necessary to continue to help reinforcing existing technologies and to introduce new paradigms for treating dental disease and restoring teeth seriously compromised by caries lesions via biomimetic and more biological operative approaches. Dental bioactive materials is arguably the latest research area in dentistry and thus the amount of new research is overwhelming. However, in this day and age of evidence based practice it important for this new information to be distilled into a practical and understandable format.

Abstract

Aging of unicellular and multicellular eukaryotic organisms is a convoluted biological phenomenon, which is manifested as an age-related functional decline caused by progressive dysregulation of certain cellular and organismal processes. Many chronic diseases are associated with human aging. These aging-associated diseases include cardiovascular diseases, chronic obstructive pulmonary disease, chronic kidney disease, diabetes, osteoarthritis, osteoporosis, sarcopenia, stroke, neurodegenerative diseases (including Parkinson’s, Alzheimer’s, and Huntington’s diseases), and many forms of cancer. Studies in yeast, roundworms, fruit flies, fishes, mice, primates, and humans have provided evidence that the major aspects and basic mechanisms of aging and aging-associated pathology are conserved across phyla. The focus of this International Journal of Molecular Sciences Special Issue is on molecular and cellular mechanisms, diagnostics, and therapies and diseases of aging. Fifteen original research and review articles in this Special Issue provide important insights into how various genetic, dietary, and pharmacological interventions can affect certain longevity-defining cellular and organismal processes to delay aging and postpone the onset of age-related pathologies in evolutionarily diverse organisms. These articles outline the most important unanswered questions and directions for future research in the vibrant and rapidly evolving fields of mechanisms of biological aging, aging-associated diseases, and aging-delaying therapies.

Abstract

The endometrium has been the subject of intense research in a variety of clinical settings, because of its importance in the reproductive process and its role in women’s health. In the past 15 years, significant efforts have been invested in defining the molecular phenotype of the receptive phase endometrium as well as of various endometrial pathologies. Although this has generated a wealth of information on the molecular landscape of human endometrium, there is a need to complement this information in light of the novel methodologies and innovative technical approaches. The focus of this International Journal of Molecular Sciences Special Issue is on molecular and cellular mechanisms of endometrium and endometrium-related disorders. The progress made in the molecular actions of steroids, in the metabolism of steroids and intracrinology, in endometrial intracellular pathways, in stem cells biology, as well as in the molecular alterations underlying endometrium-related pathologies has been the focus of the reviews and papers included.

Abstract

The way plants grow and develop organs significantly impacts the overall performance and yield of crop plants. The basic knowledge now available in plant development has the potential to help breeders in generating plants with defined architectural features to improve productivity. Plant translational research effort has steadily increased over the last decade due to the huge increase in the availability of crop genomic resources and Arabidopsis-based sequence annotation systems. However, a consistent gap between fundamental and applied science has yet to be filled. One critical point often brought up is the unreadiness of developmental biologists on one side to foresee agricultural applications for their discoveries, and of the breeders to exploit gene function studies to apply to candidate gene approaches when advantageous on the other. In this book, both developmental biologists and breeders make a special effort to reconcile research on the basic principles of plant development and organogenesis with its applications to crop production and genetic improvement. Fundamental and applied science contributions intertwine and chase each other, giving the reader different but complementary perspectives from only apparently distant corners of the same world.

Abstract

This Special Issue features recent data concerning thioredoxins and glutaredoxins from various biological systems, including bacteria, mammals, and plants. Four of the sixteen articles are review papers that deal with the regulation of development of the effect of hydrogen peroxide and the interactions between oxidants and reductants, the description of methionine sulfoxide reductases, detoxification enzymes that require thioredoxin or glutaredoxin, and the response of plants to cold stress, respectively. This is followed by eleven research articles that focus on a reductant of thioredoxin in bacteria, a thioredoxin reductase, and a variety of plant and bacterial thioredoxins, including the m, f, o, and h isoforms and their targets. Various parameters are studied, including genetic, structural, and physiological properties of these systems. The redox regulation of monodehydroascorbate reductase, aminolevulinic acid dehydratase, and cytosolic isocitrate dehydrogenase could have very important consequences in plant metabolism. Also, the properties of the mitochondrial o-type thioredoxins and their unexpected capacity to bind iron–sulfur center (ISC) structures open new developments concerning the redox mitochondrial function and possibly ISC assembly in mitochondria. The final paper discusses interesting biotechnological applications of thioredoxin for breadmaking.
